Travelling through Ecuador with a disability may be hard, but not impossible. Here are a few pointers for you:

Physically challenged: Rent a car. That is if you have someone to help you with driving. No rental car office in Ecuador offers disability ready cars. Also, it is easier to move around the city in your own car than having to pick up a taxi every time you want to move. Ecovia, Trole and Metrobus buses in Quito offer handicap spaces in their units. All of them feature ramps to enter the stations. Some stations like 24 de Mayo provide the ramp, but fail to provide ramps to cross the street.
Hearing and speech impaired individuals: There are many audio-visual elements concerning basic pedestrian signs, audio elements are not given much importance here to direct traffic. Sign language is not a common skill among the general population.
Sight impaired individuals: Moving around the city with your walking cane can be extremely dangerous. Also, help dogs are non-existent. Braille and street surface signs are nonexistent.
